Fittings like these have arrows on to show the direction of flow:

I've noticed many in my house are fitted the wrong way around.

1) Does it matter?

2) What difference does it make?

If they are over-pressured with the arrow pointing towards the source of pressure, there is a chance the "innards" will come out of the other end if its not connected to anything.

1. Does it matter. Not really other than in exceptional circumstances.
2. What difference does it make. None, the flow rate for a given pressure will be the same in either direction.

If the arrow is he wrong way it can blow the guts out the ISO valve when disconnected.
